I do a lot of trekking and so far I have been very satisfied with my Lowepro Primus. I can carry all my gear plus what I need to be comfortable (food, water, clothes, etc.)
Lowepro - Primus AW There is another version designed to carry a laptop (CompuPrimus). It also contains a visor to increase the visibility of the screen during sunny days. Lowepro - CompuPrimus AW

i didn't like the lowepro backpacks because all of the ones i had seen had the pocket on the top that wasn't much good for camera stuff or computer stuff. I ended up with a thinktank street walker HD. it says that it'll only hold a 15.4 inch laptop, but mine is an HP dv7 at 16.2 inches wide (17.3" screen size diagonally) and it fits, although just barely. it has tons of room for camera stuff too

The more I look at the backbacks though I'm not sure it's what I want now. It seems like once the camera is in there it's a little work to get them out. I'm now thinking of something like a belt pack. It's smaller, right in front, and provides quick access. I'm specifically thinking of traveling where I'll need to take the camera in and out many times. A backback doesn't seem to fit that purpose.

Kata 3-N-1. Sling or backpack, with quick side access from either side (so no worries if you're a leftie). They're now making them with and without the laptop/netbook pocket. Also a lighter-duty version they're calling the 123go.
For plain backpacks, the DR-46x series is pretty cool. They're including tripod holders, now.
