Your focus, lighting, and DOF are great. But the composition could use some tweaking. You might be able to photoshop it, but I would try to reshoot it if I were you. While I know the rule is "fill the frame, fill the frame, fill the frame" ... in this case, I think I would have backed off just a tiny little bit (not much... just a hair) and captured just enough of the background to make that wonderful DOF just a little more obvious and allow you to more strictly apply the rule of thirds. Since there is so little background, your rose appears too centered. Place the camera so it is pointing a little upward and to the right, and back off a little to grab a bit more of your background. Try that and see if you can get the rose a little more off center. Considering the shape of the rose, I would place your focal point (center of rose) in the upper right of the grid grabbing more background from the left and bottom of the shot. Good luck.
