Is this photo over-sharpened? or under? And am I using too much noise reduction so it looks soft?

It doesn't strike me as oversharpened, it just looks like an overcast day. FYI, most lenses are sharpest around f/8 and you'll get more DOF with a smaller aperture.

I suspect that you weren't harmed by the relatively wide aperture. If we assume that the car was about 40' away, your critically sharp depth of field was slightly more than 20', which should be plenty for that scene.
