Well, This is probably the wrong forum for this, but Not knowing what lens your using and your settings, it's difficult. Some lenses develop what are called Front or rear focusing issues, which means they just don't quite reach the correct focus or focus just a little too far.
No amount of Sharpening will make a poorly focused picture perfect. Just better.
Try using a Tripod and a remote shutter release next time perhaps even Mirror lock up if you really need to nail the shot.
